چکیده
The physiological disposition of l-(2-chloroethyl)-3cyclohexyl-1-nitrosourea, a highly active agent against intraperitoneal and intracranial mouse leukemia L1210, was studied in mice, rats, dogs, and monkeys with the 14C label in each of 3 positions of the molecule, the ethylene, carbonyl, and cyclohexyl moieties. l-(2-Chloroethyl)-3-cyclohexyl-l-nitrosourea is lipid soluble and, after parenteral dosage, rapidly degraded in mouse and dog plasma with 2 exponential phases. The half-life of the initial phase is about 5 min, while the 2nd phase ex tends over 1 hr. During the 6 hr after i.v. injection of ethylene-labeled 1-(2-chloroethyl)-3-cyclohexyl-1 -nitrosourea in dogs, radioactivity in the cerebrospinal fluid exceeded that of plasma 3-fold. With the label in the cy clohexyl moiety, plasma radioactivity was about 50% of cerebrospinal fluid UC levels. The drug following biotrans

Interaction of 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea (NSC 79037) with nucleic acids and proteins in vivo and in vitro.
SUMMARY The macromolecular binding of radioactivity from 1-(2-chiorethyl)-3-cyclohexyl-1-nitrosourea, labeled with ‘@ C in either the cyclohexyl moiety or the ethylene residue, was studied in the L1210 leukemia-bearing mice, in a suspension of Ll2lO leukemia cells, and during in vitro incubation with isolated nucleic acids and proteins. In all three systems, radioactivity from @ C-labeled 1-(...
متن کاملMechanism of uptake of nitrosoureas by L5178Y lymphoblasts in vitro.
The mechanism of uptake of nitrosoureas by L5178Y cells in vitro was investigated. A time course of the uptake of radioactivity on incubation of L5178Y lymphoblast with [14C]-1,3-bis(2-chloroethyl)-1-nitrosourea was linear for 30 min and then entered a plateau phase; it was markedly temperature dependent. A similar time course for cells incubated with [14C]ethylene-labeled 1-(2-chlorethyl)-3-cy...
متن کاملFormulation and biological distribution of radiolabeled 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea in an animal tumor model.
A formulation of 1-[2-chloroethyl-U-14C]-3-cyclohexyl-1-nitrosourea in ethanol:propylene glycol (1:4, v/v) was administered to rats previously infiltrated with RT6 glioma tumors (astrocytic series); the organ and tumor tissue distribution was followed at 1, 4, 12, and 24 hr postinjection. From 0 to 1 hr, rapid blood disappearance of the 1-[2-chloroethyl-U-14C]-3-cyclohexyl-1-nitrosourea was obs...
متن کاملTherapy in an intracerebral murine glioma model, using Bacillus Calmette-Guérin, neuraminidase-treated tumor cells, and 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea.
The s.c.-propagated murine glioma, GL-26, was established in tissue culture. The tissue culture line, with a doubling time of 36 hr, was used as the common source for all tumor cells. Suspensions of the tumor cells were transplanted intracerebrally in mice to produce an anaplastic ependymoblastoma. In vitro 51-Cr cytotoxicity assays did not detect any cellular immunity against GL-26 tumor cells...
متن کاملCarbamoylation of amino acid, peptides, and proteins by nitrosoureas.
Incubation at approximately physiological conditions of amino acids, peptides, and proteins with 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea or cyclohexyl isocyanate resulted in carbamoylation of the alpha-amino groups of amino acids, the terminal amino groups of peptides and proteins and the epsilon-amino groups of lysine moieties. Carbamoylation of the alpha-amino groups and the terminal ami...
